DNA barcoding analysis and phylogenetic relationships of Indian wild coffee species

Wild coffee species are the reservoirs of genetic diversity and could play a critical role in improvement coffee. However, conservation assessment wild has been largely neglected. In present study, DNA barcoding approaches were employed to assess phylogenetic relationships between five indigenous from India compared with two cultivated, four African origin. The efficacy three loci namely matK,rbcL, trnL-trnF was investigated using PCR amplification sequence characterization. intergenic spacer is highly polymorphic followed by matK rbcL chloroplast gene. Among loci, locus maximum number parsimony informative sites, whereas contains singleton variable sites. Although all contain few unique fixed nucleotides (UFNs), no individual barcode nucleotide tags for Indian that help discrimination. multilocus combinations efficient discriminating due presence SNPs specific tags. tree constructed likelihood analysis combined separated phylogeny inferred loci. Our study supports utility as useful tool identification, which can be used purposes.
